Proteinbase

The home of protein design data. An open platform by @adaptyvbio for sharing protein designs, their experimental validation and their design methods.

[Proteinbase @proteinbase]

Today we’re launching Proteinbase, a single hub for experimental protein design data. Over 1,000 novel proteins are already live, each with computational predictions, experimental validation, and the method used to design them.
Everything comes from one lab under standardized protocols, which means the results are reproducible, comparable, and include negative data that usually never gets shared.

Announcing Challenge 2 of the Anthropic x Adaptyv Protein Design Competition!

Humira, the best-selling drug ever, blocks TNF-α, an inflammation marker protein that’s overexpressed in rheumatoid arthritis and Crohn’s disease. In this challenge, the goal is to make a better binder for TNF-α that also releases selectively at pH 6.0 in the endosome of cells to improve drug half-life.

The protein must also bind both human and mouse TNF-α, making it easier to test the same design in preclinical models.

Two days left for Challenge 1 of the Protein Design Competition! You have until Tuesday, October 6 to submit your designs.

The goal of Challenge 1 is to design proteins that bind EGFR in the acidic environment of a tumour, but not at the neutral pH of healthy cells. Moreover, the proteins must bind to both the human and the mouse variant of EGFR to support easier preclinical testing.

Submit your designs on @proteinbase now: https://t.co/qRvrOYMJs0

The Protein Design Competition is back, bigger and better than ever thanks to our partnership with @AnthropicAI

The competition will feature five challenges, each focused on a specific disease or biological mechanism. Compared to previous competitions, it will be a big step-up in complexity and scale to push the boundaries of AI-driven protein design.

The competition is open to everyone and free to enter. It will feature 3 tracks:

- Track 1 is aimed at expert protein designers, with up to 20 teams to be selected.

- Track 2 is targeting life science academics and industry researchers.

- Track 3 is open to everyone from tech enthusiasts to high-school students.

A big thanks to @Modal for contributing compute for protein design and to @TwistBioscience for contributing the DNA for the experimental validation!

We raised a $40M Series A to build the automated lab for agentic biology.

Last week, we shared the work we did with @Anthropic: Claude designed proteins, sent them to our automated wet lab, and got real experimental data back.

We believe this is where biology is going: AI agents designing experiments, running them in automated wet labs, learning from the results and iterating.

But AI can only move as fast as the experiments behind it. To use the potential of AI to cure all diseases, we need to build high-throughput wet lab infrastructure: a “biological gigafactory”.

That’s exactly what we’re doing at Adaptyv.

Over the past year we’ve grown our lab throughput by over 5x and onboarded more than 100 customers, ranging from bio AI labs like @chaidiscovery and @boltz_bio to pharmas like @Roche and @novonordisk to dozens and dozens of new startups that use AI to radically speed up drug discovery.

What happens when you let frontier LLMs design proteins, and then synthesize and test them in a wet lab?

We ran a protein design competition with @muni_bio where AI agents competed against humans to design molecules that bind TREM2, a key receptor linked to Alzheimer’s.

Results: GPT 5.2 and Grok 4.1 both placed in the top 5, with molecules showing strong binding to TREM2 when tested in our lab.

Negative Results Bundle 😅

We just published 3 new collections on Proteinbase, and they're mostly negative results:

· Boolean Biotech's (@btnaughton) VHH Competition (6 VHHs vs MBP)

· @EvolvedTechInc Hackathon (80 designs vs CD16a/CD16-2)

· PD-L1 FoldCraft by @KhRRustamov (35 de novo vs PD-L1)

Clean non-binder data is still quite rare and useful, especially for training models. All open under ODC-ODbL.

🧬 We picked the 300+ designs going to the wet-lab for the @gembioworkshop x @adaptyvbio RBX1 Binder Design Competition.

12000+ submissions from 180+ designers. About 6x the volume we had expected.

Expression and binding affinity measurements are now running. Results will be presented at the @gembioworkshop workshop at @iclr_conf 2026 in Rio 🇧🇷

Submissions for the RBX1 protein binder design competition close tonight at 00:59 CET (March 27).

Quick recap: Design protein binders against RBX1, a cancer-relevant E3 ligase subunit. Half disordered, RING domain with zinc ions, making it a real design challenge.

300 designs will be experimentally tested in the @Adaptyv lab. Results at @iclr_conf 2026 in Rio. Open to everyone.

Submissions are open for the GEM x @adaptyvbio RBX1 Binder Design Competition 🧬

We're partnering with the GEM Workshop at ICLR 2026 for a new protein design challenge. The target is RBX1, a 108-amino acid component of Cullin-RING E3 ubiquitin ligase complexes, which control degradation of roughly 20% of all cellular proteins. In many cancers, this protein is overexpressed and degrades important tumor suppressing proteins. Disrupting RBX1's function with a de novo designed binder could slow down cancer cell proliferation.

But RBX1 is a tricky design problem. Half the protein is an intrinsically disordered N-terminus so it has limited use for structure-based design methods. The part you actually need to bind, the E2 recruitment surface on helix α2, sits within a RING domain with an usual fold that is shaped by three coordinated zinc ions.

How it works:
- Submit up to 100 ranked binder sequences (≤250 aa) with a design method description on Proteinbase till March 26
- An expert panel from the GEM workshop selects 300 designs based on novelty and originality
- We test all 300 for expression and binding affinity in the @Adaptyv Bio lab
- Experimental results will be released April 26 at ICRL 2026 in Rio de Janeiro and open-sourced on Proteinbase

Prizes: $1,000 for the best binder, $100 for the runner-up, and co-authorship on the results paper.

We just added new protein card highlights to better show de novo designs on @proteinbase!

We're checking all sequences against a bunch of databases via MMseqs2 search to ensure that the designs are not similar to already existing proteins.
