Raising NAD levels reduces inflammation in humans.
At least 8 randomized controlled trials show that ~1 gram per day of nicotinamide riboside (NR) lowers key inflammatory cytokines, including IL-6 and TNF-α.
NAD depletion is a hallmark of chronic inflammatory states, from respiratory infections to metabolic disease to unhealthy aging. In many of these conditions, inflammation itself disrupts NAD metabolism, creating a vicious cycle where inflammation lowers NAD, and low NAD impairs the body’s ability to resolve stress and repair damage.
NAD is required for DNA repair, mitochondrial energy production, sirtuin signaling, and cellular antioxidant defenses. When NAD drops, these systems suffer.
@CharlesMBrenner explains that restoring NAD with precursors like NR may help break that cycle by supporting the body’s natural repair, detoxification, and anabolic processes.
Importantly, these effects have been observed in randomized human trials, not just animal models.

Higher body weight elevates risk in two ways. First, every extra pound adds up to 4x as much force to the knee joint.
