A randomized phase 2 trial evaluated talazoparib plus enzalutamide 🆚 enzalutamide 💊 alone in high-volume metastatic hormone-naive #ProstateCancer
➡️ Although combination therapy achieved a 73% PSA response rate at 12 months, superiority over monotherapy was not established.
➡️ Grade ≥3 adverse events were more frequent with combination treatment, including two fatal myeloid neoplasms.
✅ Tumor analyses revealed androgen receptor inhibition altered gene expression without impairing functional homologous recombination, highlighting challenges in identifying patients most likely to benefit.
