A new biocontainment strategy to prevent engineered cells from growing outside the lab.
About 775 proteins that are "essential" to yeast growth were tagged with a small protein, called a degron. This degron destabilizes the proteins in the absence of estradiol, but not in its presence.
When one essential gene, called SPC110, was tagged in this way, fewer than 1 in 100 million yeast were able to survive in the absence of estradiol after 10 days.
In the lab, engineered yeast that are grown in media containing as little as 100 nM of estradiol are able to grow just as quickly as wildtype cells.
This biocontainment method is "mechanistically orthogonal to previously reported genetic biocontainment systems," meaning it can probably be coupled with other approaches to make something even more robust.
