Agree @PTarantinoMD, this trial has important (& very concerning) limitations. That said, the association is reproducible across >15 independent studies (some also biased, sure), and is supported by preclinical data in @CellCellPress (DOI: 10.1016/j.cell.2024.04.015) showing circadian regulation of tumor-infiltrating CD8⁺ T cells, with diurnal peaks in cytotoxicity and PD-1/ACT efficacy—implicating the endogenous clock as a biologically relevant modulator of immunotherapy response.
