Bit late, but first paper of 2025. 🧬Enhancing #singlecell#transcriptomics: Our recent study introduces an interposed anchor strategy to improve gene expression profiles in droplet-based single-cell sequencing analyses. https://t.co/owRLEKcMFu
Latest #BioRxiv on scRNA-seq reveals critical issues with droplet-based beads: Truncation compromises UMI identification, leading to inaccuracies in differential expression analysis. A must-read if your using 10X Genomics or dropseq in your research. https://t.co/5krBizVnnm
I am so thrilled to see that our paper on "improving molecular counting with homotrimer UMIs" out NOW in Nature Methods @naturemethods! Details of the entire analytical approach for the multiplexing and demultiplexing of homotrimer UMIs are available NOW!
Out today: from @AdamCribbs and colleagues, a method utilizing homotrimeric nucleotide blocks achieves accurate counts of RNA molecules in bulk and single-cell sequencing studies.

Falling behind: postdocs in their thirties tire of putting life on hold.

A real goodbye to my old lab as my last co-authoring paper @thewulab is officially out now. It's sheer joy snorkelling to catch sea urchins as specimens and formulating a protocol that helped with this project during my self-doubting times. Been a great 3 yrs. Thx @thegellybear!
What's more awesome than finally getting a study published? Getting it published with your good friend as the co-corresponding author! @drpluteus thank you for accompanying me on my first foray into the ocean! (And hope this is the first of many...)

If anyone is working with sea urchins and is in need of a protocol to extract high MW genomic DNA from sea urchin tube feet for long-read seq, please reach out :))

2) We demonstrated that through trimerisation of the UMI random sequence (ATC —> AAATTTCCC) complemented with our computational toolkits, we were able to push the transcript quantification accuracy to >99%.

New work from new lab:
1) We revealed that UMI does not accurately correct transcript quantification (as opposed to what people thought), and that PCR error but not sequencing error is the source of UMI inaccuracy regardless of sequencing platforms (Illumina, PacBio & Nanopore).
New preprint from our group. Q: How accurate is bulk and single-cell RNA sequencing? A: Not as accurate as we expected. In fact, at the single-cell level, PCR bias has a significant effect. We quantify this experimentally. A 🧵 https://t.co/xXAMe1ajGB #SingleCell
Congrats to @thewulab and really happy and honoured as a co-author to see this #singlecell dual-omics technology scONE-seq finally making its way out of the womb after almost two years since submission!
Took a while, but it's finally published! #singlecell#ScienceAdvancesResearch#HKUSTResearch scONE-seq: A single-cell multi-omics method enables simultaneous dissection of phenotype and genotype heterogeneity from frozen tumors https://t.co/R8Q5vH1hvX
Finally….. my first PhD pub is out!!!!!! Very cool action from the host in restricting replication of a Herpesvirus….BUT the virus outsmart it🦠

The finding: single-cell analysis reveals detrimental effects of insomnia on immune cells.
The irony: the main cause of my insomnia is single-cell analysis

The science journal publishing industry is a scam because we give publishers all of the content FOR FREE, we provide most of the labor that adds value FOR FREE, and we give journals $10b/year to “cover their costs”, we don’t even get universal access to the final product.
Check out our freshly published rRNA depletion method (scDASH) @thePeerJ which made profiling of total RNA achievable in single cells. All credit goes to @thewulab!
