Metformin for patients with metastatic prostate cancer starting androgen deprivation therapy: a randomised phase 3 trial of the STAMPEDE platform protocol

In the STAMPEDE phase 3 trial, researchers evaluated whether adding metformin to standard of care could improve survival in non-diabetic patients with metastatic hormone-sensitive #ProstateCancer . Among 1,874 participants, no significant improvement in overall survival was observed with the addition of metformin (median OS: 67.4 vs. 61.8 months; HR 0.91, p=0.15). While metformin was associated with more gastrointestinal grade ≥3 adverse events, it had an otherwise expected safety profile. Notably, metformin significantly reduced the adverse metabolic effects typically associated with androgen deprivation therapy.

These findings suggest that while metformin does not improve survival, it may offer metabolic benefits in this patient population.
