@lopantano@lopantano by default Cell Ranger only uses 16 GB for the ALIGN_READS stage, someone had to manually set it to 220 GB and so override the default when they ran mkref. The 64 GB you found also seems too high, the _jobinfo file can help you refine that estimate.

I am happy to announce Rust-HTSlib 0.21 and Rust-Bio 0.22. Rust-HTSlib contains many small usability improvements (a lot of them implemented by @10xgenomics). Rust-Bio provides a comprehensive HMM implementation (thanks to Manuel Holtgrewe). Great work by a growing community!
