Started a BRAF Colorectal Cancer Public Library group on Facebook. So many significant trials, research findings, changes to NCCN guidelines, etc. in the last 2 years. Gathering high level research/interviews into 1 place seems wise and useful. #BRAFCRC https://t.co/1FjFjY9AOU
By showing a near-doubling of overall survival already in the second-line setting, daraxonrasib is an example of a “transformative therapy” that makes conducting future, randomized clinical trials more challenging logistically and ethically, Pazdur said.
“If you’re going to do a first-line trial, you have the potential of losing equipoise,” Pazdur said. “People are going to want the drug. Let’s face it, if you have a fatal disease like pancreatic cancer and you are going to die from that disease, you do not want to go on the [control] arm.”
Pazdur proposed alterations to $RVMD's current Ph3 trial in first-line pancreatic cancer that could work to expand the drug's use and get it to patients faster.

I also spoke w/ RevMed CEO Mark Goldsmith about the same.
Finally got our first #pancreaticcancer patient onto #daraxonrasib through @RevMedicines@FDAOncology Expanded Access Program. Turned into an unplanned bring-your-daughter-to-work day to get same-day consent to start therapy. Fingers crossed!
#ASCO26 reinforced what we know: RAS-targeted therapies are reshaping cancer treatment. Our Phase 3 RASolute 302 results demonstrate meaningful progress for patients. Excited about what's ahead.
Great session! With lots of hopeful collrectap cancer research for future ASCOs! Better patient outcomes and clinical guidelines for care
Wonderful way to end #asco26@ASCO
🙏🙏 @ASCO@Betzabe100@KristenCiombor We had a wonderful time creating this CRC IO Education Session; case debate/voting/Q+A too! Thank to Stacey Cohen and our moderator, Chris Lieu a ASCO-FASCO pro! Thank you ++ to the ASCO AV team who were so helpful+patient with us! 🙏🙏🤝🤝
The work is far from finished. As excitement builds, researchers are already focused on the next questions: how to achieve more durable responses and understand resistance to help further improve outcomes for patients—questions at the heart of our collaborative daraxonrasib study with Revolution Medicines, which is designed to understand not just whether this therapy works, but how and why it works in patients.
Daraxonrasib, the first KRAS drug for pancreatic cancer, is now available through an expanded access program, but your physician must apply for you. https://t.co/cUA7KU8msU
4. What's next for Daraxonrasib? More trials!
1st line in metastatic pancreatic cancer, and after surgery in resectable disease; in lung cancer; and in other solid cancers, like colorectal
MIRACLE-2: RT to primary/mets -> chemo + tislelizumab in MSS unresectable met rectal ca (N=50): 68% ORR & median OS 23 mo.
Early, single-arm data, but ~1 in 5 pts reached NED.
Suggests RT + systemic + PD1 blockade could overcome immune resistance in MSS mCRC. #ASCO26@OncoAlert
PanCAN's Chief Scientific and Medical Officer, Dr. @ABerkenblit, shares, "I know that in the years to come, we'll look back on this year's #ASCO26 meeting as marking the beginning of a new era for patients with pancreatic cancer. As a long-standing American Society of Clinical Oncology (ASCO) member, it was a privilege this year to attend alongside my colleagues from Pancreatic Cancer Action Network, knowing the pivotal role PanCAN played to get us to this historic moment, and that the community is looking to PanCAN to help us all make good on the promise that RAS therapies hold for patients with pancreatic cancer.
Unprecedented indeed are the Phase 3 data for #daraxonrasib, a RAS-targeted therapy developed by Revolution Medicines and presented by Dr. Brian Wolpin. These highly anticipated data were received with wild enthusiasm — a standing ovation! — from the ASCO community. Overall survival and progression-free survival were ~doubled, and overall response rate was ~tripled with daraxonrasib compared with chemotherapy in patients with previously treated metastatic pancreatic cancer, with all subgroups benefiting. Importantly, quality of life and pain were improved, so that patients not only live longer, but also feel better — just what we want from new therapies. Optimal management of side effects, including rash and mouth sores, is paramount to maximize the benefit — and PanCAN Patient Services is ready to embrace the educational needs!"

@DrSukeshiArora@RevMedicines The first shipments went out last week.
The paperwork is streamlined through cancer centers.
I dont know how it is/isnt working for community oncologists.
Great question for @RevMedicines
#ASCO26 | Abstr 3510
CRDF-004: rPh2 Onvansertib + Chemo + Bev in 1L RAS-mutated mCRC
Promising ORR and PFS HR, but results should be interpreted with caution due to the small sample size. Ph3 is planned.

@kathmacphail@DrHBurstein also -
the distinction between a "standing ovation"
and
"an interrupting standing ovation"
Also - great point in chat re need for patients to understand more of these concepts.

Agree with @kathmacphail - that was a great conversation!
The application of daraxonrasib IN THE CLINIC will provide so much info for clinicians to move forward.
@DrHBurstein's important point about whether enrollment in a first line trial is even possible now (loss of equipoise).
