Everyone's racing to engineer T cells for solid tumors. One of the most striking pediatric brain-cancer results this year came from T cells that weren't engineered at all.

Children's National just published a Phase 1 in Nature Medicine: multi-antigen T cells targeting WT1, PRAME and survivin — non-gene-modified, delivered by simple IV — in children with DIPG and relapsed CNS tumors. DIPG is a death sentence; median survival is roughly a year.

The primary endpoint was safety, and it cleared. The signal underneath is the story: three children in the relapsed arm remain disease-free with no further treatment — the longest past four years — including one complete remission. And it's a non-engineered approach, built to sidestep the neurotoxicity that makes CAR-T so hard to run in the brain.

The read for markets: the only public company built on this exact modality is Marker Therapeutics. Its MT-601 stacks those same three antigens plus three more, and it's aimed squarely at patients who've already failed CD19 CAR-T.

Single-target CAR-T has the spotlight. This is the counter-thesis worth watching.
