Paper identifies (3) features of better T-cell fitness: 1. Restrained chronic #TCR signalling 2. Avoidance of #NFAT→#TOX/#NR4A exhaustion 3. Preservation of #BACH2/memory-state programs These align very well w/ effects of selective #ITK inhibition and paper somehow misses this connection. Paper explicitly argues for an optimal TCR signalling window, avoiding chronic/high-intensity TCR signalling which is a core exhaustion driver. Concept fits #soquelitinib particularly well as it allows this signal modulation. I modified image from paper to show central role ITK plays in Signal 1 TCR signaling and how it can effect key downstream pathways: NF-κB, AP-1, NFAT.
