8 years ago a drug called Venetoclax comarketed by Abbvie and Genentech was first approved for CLL ,and then SLL and AML were added to its label later. It was tested because it inhibits BCL-2, which is a cell survival protein with the thought that cancer cells would die more often. And indeed it has had a great response on these leukemias. Then late last year an increasingly highlighted and studied article was published by Zhao et al (Cancer Discov (2023) 13 (11): 2448–2469) that astonishingly found that BCL-2 inhibition-related increased apoptosis is NOT and never has been the primary reason that this drug works so well in leukemias. The MAIN reason is that BCL-2 is a checkpoint that RESTRAINS DENDRITIC CELL FUNCTION, and therefore BCL-2 inhibition results in activated dendritic cells. Now does it make sense why cancers love to express BCL-2? They’re trying to stop their dendritic cells from doing their jobs. When combined with PD-L1 inhibitors Venetoclax is even more potent. Now just imagine what would happen when it is combined with $NWBO DCVax and PD-L1 inhibitors like Keytruda… Talk about the end of cancer as we know it!!!
