Why Iโm bullish on $RVMD and RAS(ON) inhibitors.
Most people looked at Revolution Medicineโs daraxonrasib data this weekend and saw a pancreatic cancer breakthrough.
And to be fair, it is.
Doubling median overall survival in metastatic pancreatic cancer is the kind of result that earns a standing ovation at ASCO.
But I think the bigger story is what the data may say about Revolution Medicine itself.
For decades, RAS has been one of the most important targets in cancer biology. Mutations in the RAS pathway are found in roughly 25-30% of all human cancers, including pancreatic, lung, colorectal, ovarian, and many blood cancers.
The problem was that RAS was considered undruggable.
Scientists knew it was driving cancer. They just couldnโt figure out how to shut it off.
Thatโs the opportunity Revolution Medicine was built around.
Rather than developing a single drug for a single cancer, the company built a technology platform designed to target RAS proteins in their active, cancer-driving state. Their approach uses what they call RAS(ON) inhibitors, a new class of medicines designed to directly engage activated RAS signaling.
Daraxonrasib is one product of that platform.
The important question isnโt whether daraxonrasib can become a successful pancreatic cancer drug.
The important question is whether these results represent the first large-scale validation of Revolutionโs broader approach to targeting RAS-driven cancers.
Because if the underlying technology works, the opportunity extends far beyond pancreatic cancer.
It potentially extends into lung cancer, colorectal cancer, and a range of other tumors where RAS mutations act as a core disease driver.
The best biotechnology companies donโt create a single blockbuster.
They create a repeatable engine for discovering them.
Genentech did it with antibodies.
Vertex did it with CFTR biology.
Moderna did it with mRNA.
What investors are trying to determine now is whether Revolution Medicine has built a similar engine for one of the most important pathways in oncology.
Itโs still early. Resistance mechanisms will emerge. Additional trials will need to confirm the breadth of the approach.
But this weekend may ultimately be remembered as something bigger than a pancreatic cancer breakthrough.
It may be remembered as the moment the oncology community started believing that RAS, one of cancerโs most important biological engines, could finally be systematically targeted.
