@Professor_Tong Thanks @geewhiz_ephys! Yep you need to cryoprotect - we would perfuse with calcium free Tyrodes then fixative of choice. Full laminectomy to dissect out. If well perfused then 30% sucrose for at least 24hours before making blocks and freezing!

@ohambiguity My PhD lab (and my PIs PhD and postdoc lab) first perfused with calcium free tyrodes, then for fixative used 4% paraformaldehyde and 0.2% picric acid in 0.1M phosphate buffer, pH 6.9. Stable for 3 weeks at room temp.
