@hsalis I think they're either using bacteria to do nick repair, but they do mention use of PNK in one section of the methods:
"The ligation reaction (20 μl total volume) consisted of 17 μl assembled fragments, 2 μl of 10× T4 DNA ligase buffer and 1 μl polynucleotide kinase."
@ATinyGreenCell Gotcha. I've had them make ultramers with 20nt at 50%gc and then 100T in a row for adding polyA tails to mRNA templates, but agree, not sure about 0% GC.

@ATinyGreenCell That would depend if there is a cryptic promoter upstream I guess. I'm assuming that atg is part of the -10 region of the promoter, so probably don't have any transcription from that point.
@ATinyGreenCell There is an atg upstream, is that in the other strength promoter versions? Any chance this one is making a small peptide that is toxic? The deletion is removing a stop codon in frame to the atg in the promoter.

@breakliquid Problem of wanting to use thin walled plastic to get better heat conduction, but makes them flimsy. I solve it by putting a strip of tubes at each side of the well block. Decreases pressure on your tubes.
