CAR-T without manufacturing?

A new Nature highlight shows we may soon generate CAR-T cells directly inside the body 🧬

⚙️ In vivo CRISPR strategy
• EDV → delivers Cas9 + gRNA to T cells
• AAV → delivers CAR template
• Targeted insertion into TRAC locus

📊 What they saw (mouse models)
• 5–20% of T cells became CAR+ in ~2 weeks
• Robust CD19+ B-cell depletion
• Leukemia models → complete responses across treated mice

💡 Why this matters
• Removes ex vivo manufacturing bottleneck
• Moves toward off-the-shelf CAR-T
• Potential to scale cell therapy globally 🌍

⚠️ But still early
• Preclinical only
• Off-target editing + safety concerns
• Human efficacy remains uncertain

🚨 Physical Activity and Cancer Mortality in Survivors.

Pooled analysis of 6 cohorts (n=17,141) shows post-diagnosis moderate to vigorous physical activity is associated with lower cancer mortality across several cancers.

Even low activity levels reduced mortality in bladder (HR 0.67), endometrial (HR 0.62), and lung cancer (HR 0.56). Meeting activity guidelines after diagnosis improved outcomes in lung (HR 0.58) and rectal cancer (HR 0.51), even in previously inactive patients.

PD-L1 and TMB remain imperfect predictors of response to ICIs in NSCLC. Emerging biomarkers include genomic alterations (KEAP1, STK11, SMARCA4), tumor-infiltrating lymphocytes, metabolic pathways (IDO, adenosine), and blood-based markers such as ctDNA and inflammatory signatures. AI-driven composite models may improve prediction.
