Calico Life Sciences announced today that their eIF2B activator investigational drug, Fosigotifator (ABBV-CLS-7262) was selected for the FDA START Pilot Program. This START Pilot Program was established to further accelerate the development of novel drug and biological products for rare diseases.
In this particular study, Fosigotifator (ABBV-CLS-7262) will be investigated in participants diagnosed with Vanishing White Matter (VWM) disease, an ultra-rare progressive leukoencephalopathy. VMN is caused by variations of the eIF2B subunit, an essential enzyme in the integrated stress response (ISR). When ISR is activated in a normal state, global protein expression is reduced (controlled) to enhance the expression of a signature set of stress response genes. But in VMN, the function of eIF2B is reduced, triggering chronic activation (uncontrolled) of ISR, leading to a loss of protein homeostasis (a hallmark of aging).
Other research groups have shown that ISR activity also increases with age and are currently studying the connection between ISR and brain aging. We've learned a lot about aging biology through rare diseases and hope to continue learning through these channels.

The United States is facing a shortage. An organ shortage. With approximately 120,000 Americans languishing on waitlists, and only 45,000 transplants performed annually, we have reached a critical juncture if we intend to solve this problem.
In light of this, the Advanced Research Projects Agency for Health (@ARPA_H) has recently unveiled its Personalized Regenerative Immunocompetent Nanotechnology Tissue (PRINT) program to combat this issue.
By utilizing cutting-edge bioprinting technology, PRINT aims to craft personalized organs on demand in order to significantly reduce wait times and improve access to life-saving treatments, as well as eliminate the need for lifelong immunosuppressive drugs.
