@inobrodib We used this information to develop second generation molecules of CSS1477 with enhanced selectivity. Our study provides a new foundation for designing domain specific inhibitory small molecules to maximize the sensitivity of cancer cells to therapy.
We describe how Group 3 medulloblastoma cells were especially sensitive to EP300/CBP BRD inhibition by @inobrodib (CCS1477), as compared with HAT inhibition. Using structural biology, we determined the mechanism by which CCS1477 binds selectively to the BRD of EP300 and CBP.
