Key points:

6/7: Breakthrough infections (after 3rd vaccination) also induced a short-lived CD8+ T cell effector response, which was compareable in phenotype and dynamics to a 4th vaccination.
5/7: Importantly, this memory pool was already formed after the 2nd vaccine dose and shows limited phenotypic changes after boosting, regardless whether specific memory subsets (TSCM & TCM) or more comprehensive measures to define memory cells (CD127+ & BCL-2) were analyzed.
4/7: Further analysis revealed the presence of classical memory cells throughout the effector phase upon boost vaccination, suggesting effector responses are built on top of a stable memory pool.
3/7: Phenotypically, the effector responses induced after each vaccination are indistinctive. Importantly, CD8 T cells with high BCL-2 expression were maintained upon boost vaccination.
2/7: Spike-specific CD8+ T cells are efficiently activated (CD38+, Ki67+, T-bet high) and expanded by a 3rd mRNA vaccination. This boosting effect lasts approx. 2 months.
