Through tirelessly efforts, our graduate student @cheni_hsu published a manuscript in @STARProtocols to reconstitute enzymatic activities for ultra-large (>300kDa) histone methyltransferases NSD1 and SETD2. To our best knowledge, this is the first time their full-length forms have been reconstituted for enzymatic assays. These advances will allow unprecedented functional interrogations towards them, especially probing into the large unstructured and potentially disordered regions, to better understand their roles in basic epigenetic mechanisms and human diseases.

I'm thrilled to share my lab's first manuscript identifying that paraspeckle protein NONO regulates active chromatin through allosteric activation of NSD1 on BioRxiv: https://t.co/LKMNwMrTCV
As a baby PI who started in a literally empty building, we weren't able to do biochem until a year after. Very proud that my PhD student @cheni_hsu pulled this through within 2.5 years!
In this study, we uncovered a novel allosteric mechanism of NSD1 central to active chromatin formation and early development. Surprisingly, we identified NONO allosterically stimulates NSD1 through the aromatic pocket of PWWP2 domain in NSD1. NONO deficient stem cells partially phenocopies the defect of neural differentiation in NSD1-null cells. Our work has several key implications in the interplay between paraspeckles and active chromatin, the development of possible allosteric inhibitors for NSD1, and understanding neurodevelopmenal disorders in NSD1- and NONO-mutant patients.

@UMassChan @HHMINEWS @theNASciences@theNASEM@aaas @WUSTL I honestly believe that he had 20 more years of good science in him. He even just submitted an @NIH grant proposal last week! He inspired me to be a nice person, but he inspired me in many other ways as well -- to work hard and to think hard.
@UMassChan @HHMINEWS @theNASciences@theNASEM@aaas @WUSTL He eventually softened on genomics. He became a major practitioner of genomics. He softened on the value of taking on scientific leadership positions. He was fun to see him flip his attitudes about things he once criticized me for!
@UMassChan @HHMINEWS @theNASciences@theNASEM@aaas @WUSTL He was one of those molecular biologists who had reservations about the Human Genome Project. He was bothered that I was going to “waste my career” on it. He feared I was killing my research career with the HGP!
My older brother, Michael R. Green, passed away suddenly and unexpectedly at the age of 69 late last week. This is a huge loss -- for his wife Maria, for @UMassChan, for me, for science.
Research opportunities at CHOP and Penn! We are seeking motivated college graduates for a lab research technician position in immune system/cell engineering! candidates with cell culture and animal handling experience are preferred. Check our lab site: https://t.co/mhicINdtNz
It is with heavy hearts that we announce the passing of our beloved colleague and friend, C. David Allis. Aside from his groundbreaking contributions to biomedical science, David will be remembered for his warmth, humility, and relentless optimism.
