Introducing 🪩 DISCO: Diffusion for Sequence-structure CO-design
14 rounds of directed evolution and over a year of wet lab work. That's what it took to engineer an enzyme for selective C(sp³)–H insertion, one of the most challenging transformations in organic chemistry.

We found that many bacterial species use exogenous peptidoglycan fragments - released by lysis of neighboring cells - as a general danger signal, triggering a danger response that protects bacteria against many dangers: biofilm formation.

After a lot of effort, and multiple rounds of improvement, this work, forming the main part of my Master’s Thesis at IISc in the lab of Prof. Dipankar Nandi @DipankarNandi97 , is finally out.

We developed novel interfacial probes as a diagnostic for membrane dynamics in bacteria, and discovered a new paradigm of antibiotic damage - cytoplasmic membrane thinning, in work spanning across 3 labs, over the past 2 years.

All in all, we characterised a different way of looking at antibiotic resistance - to look at alterations (or lack thereof, under resistance) in the bacterial membrane upon antibiotic action, with potential for diagnostic usage.
