4. An in-vitro surrogate for a long-term clinical outcome 🔮 Our in-vitro readout of gene marking in the HSC cluster associated with the vector copy number later measured in patients ~1 year after gene therapy!
3. My personal highlights: we caught stem cells self-renewing ex vivo.
Two independent clonal-tracking methods recovered the SAME clonal mark in different mice given aliquots of one culture.
👯♀️ What this means: 1 HSC split into 2 daughter cells that retained stemness!
2. First we characterised the problem at single-cell resolution.
By scRNA-seq we can zoom in on molecularly defined HSC clusters, focusing on specifically the rare cells that truly matter!
