From bladder cancer to prostate and kidney cancer, several GU oncology studies are already generating strong attention ahead of @ASCO 2026.
To identify the trials most likely to shape discussions at the meeting, we asked leading AI models to rank the most anticipated GU cancer readouts.
There was particularly strong consensus around EV-302, PROTEUS, RAMPART, and TALAPRO-3 highlighting continued momentum for ADCs, radioligand therapy, immunotherapy combinations, and intensified hormonal strategies across GU cancers.
The list underscores key themes shaping the field:
▶️ Expansion of ADC and immunotherapy combinations in urothelial cancer
▶️ Continued evolution of AR-targeted and PARP-based approaches in prostate cancer
▶️ Growing momentum for radioligand and PSMA-targeted therapies
▶️ Increased focus on treatment optimization and de-escalation strategies
▶️ Novel approaches in RCC, NMIBC, and metastatic hormone-sensitive disease
Notable studies include:
• EV-302 evaluating enfortumab vedotin + pembrolizumab in metastatic urothelial cancer
• PROTEUS assessing apalutamide intensification in high-risk localized prostate cancer
• TALAPRO-3 advancing talazoparib-based combinations in HRR-altered mCSPC
• PSMAddition exploring 177Lu-PSMA-617 in PSMA-positive mHSPC
• RAMPART evaluating adjuvant durvalumab strategies in renal cell carcinoma
Together, these studies reflect how GU oncology continues to move toward more biomarker-driven, targeted, and combination-focused treatment strategies heading into ASCO 2026.

Ahead of the @AACR 2026, we asked leading AI models to forecast outcomes for high-impact oncology trials ahead of the official AACR presentations.
We analyzed 22 trials, generating 40 predictions across key endpoints, along with 10 predicted Kaplan–Meier curves to visualize how outcomes may separate over time.
Each forecast includes hazard ratios, statistical significance (p < 0.05: Yes/No), median survival estimates, and AI-modeled Kaplan-Meier curves across tumor types.
Highlights:
Strong signals
IMCgp100-202 (Uveal Melanoma): Tebentafusp expected to sustain long-term survival benefit
▶️ AI consensus OS HR: 0.65
NCT05204628 (NSCLC): Dirozalkib projected to significantly improve PFS vs crizotinib
▶️ PFS HR: 0.46
NCT05969171 (Pancreatic Cancer): Surufatinib + chemotherapy shows promising PFS benefit
▶️ PFS HR: 0.67
Mixed / limited benefit
PRAGMATICA-LUNG (NSCLC): Ramu + pembrolizumab expected to show minimal OS improvement
▶️ OS HR: 0.96 (not significant)
Early-phase momentum (ORR signals)
Morpheus-Liver (HCC):
▶️ ORR: 41%
JS207-based combinations (HCC / mCRC):
▶️ ORR: up to ~50%
ZL-1310 (DLL3 ADC):
▶️ ORR: ~43%
Across trials, the AI consensus points to durable benefits in select targeted and immunotherapy approaches, strong early signals in next-generation combinations and ADCs, and continued variability in late-stage outcomes.

Ahead of the European Lung Cancer Congress 2026, we asked leading AI models to predict outcomes for key lung cancer trials. Now that the data is out, we compared those forecasts with actual results presented at ELCC 2026.
Important notes:
• Exact matches were suspected of data leakage due to being overly precise, so they were removed.
• Additional trials were also excluded because median values were not reported.
Where AI got it directionally right
HARMONi (IC PFS)
Ivonescimab + chemo showed strong benefit as predicted
▶️ AI: HR 0.36 | Actual: HR 0.53 (both significant)
LATIFY (OS)
Limited benefit correctly anticipated
▶️ AI: HR 0.98 (NS) | Actual: HR 0.90 (NS)
OptiTROP-Lung03 (OS)
Clear OS improvement captured
▶️ AI: HR 0.59 | Actual: HR 0.63 (both significant)
Where AI underestimated benefit
TOP (PFS)
Osimertinib + chemo outperformed expectations
▶️ AI: HR 0.65 | Actual: HR 0.44
▶️ Median PFS separation wider than forecast
Mixed alignment
NCT04143607 (OS)
Directional signal close, but significance not achieved
▶️ AI: HR 0.79 (NS) | Actual: HR 0.78 (NS)
Takeaways
• Across trials, AI forecasts:
• Captured directionality well (benefit vs. no benefit)
• Struggled with magnitude of effect (especially strong wins)
• Performed best in established mechanisms (IO, chemo combos)
• Showed more variance in targeted therapy settings
AI is increasingly reliable for signal detection, but less precise on effect size and separation, especially in high-impact trials.
