New manuscript is on BioRxiv! We (+@StephCragg) found that ChIs (via nAChRs) profoundly, continuously, and dynamically DEPRESS DA release, and dissociate cell body activity from striatal DA release, by preventing subsequent depolarisation of DA axons.

@trose_neuro And all the artifact (e.g. ~20 % decrease during REM) of widefield imaging data has been corrected using 405 nm excitation (approximately isosbestic point of 5-HT sensor). The effectiveness of this correction has been validated with membrane-tethered EGFP expressing control mice.
@blsabatini Curious about isosbestic points for green sensors like GRAB-DA, dLight, etc. @yulonglilab commented in a thread about their (green) 5-HT sensor that isosbestic is 405 nm - is this different across all the sensors, even within the GRAB family?

@LauraGrimaNeuro @NicTritsch @yulonglilab@NYULH_Neuro@LauraGrimaNeuro all mice had a minimum 5 days habituation to the behavioral setup (they usually get used the the wheel by the ) and then we recorded from them in the weeks following. Not sure if that answers the whole question so feel free to DM me if not!
