The value of 3.7 kDa was based on a literature source but it is possible we misunderstood the weight. We'll double check it. As for yields, our calculations are based on vg production so a heavier weight shouldn't affect usable AAV vg yield, only final AAV weight.

One of the big challenges to the continuous upstream was deciding how to manage the large volumes produced and then purify them in batch form. We decided to use two columns for our upstream portion and fill them as product is drawn to save time and reduce complexity.

Some amount of dye appears in the rinse water each wash (except for prototype 1). Does this mean more dye is washed out each time? Is there a way to measure total dye loss? Was the wash volume controlled too?
