💊 NALIRIFOX in mPDAC (real-world, N=80) 🔗 https://t.co/QEbwk0wfTz
⏱️ TFS 5.7 mo · OS 11.2 mo (≈NAPOLI-3) ✅ DCR 57.5% ⚠️ Gr 3-4 AEs 22.5% 🧬 HRRm → OS 16.2 vs 11.1 mo 🔬 KRAS G12D/V → worse OS 📌 Confirms efficacy, safety & role of molecular stratification
💊 NALIRIFOX in the real world for mPDAC
🔎 Multicenter retrospective (Austria, N=80) confirms findings from NAPOLI-3 in daily practice:
👥 Population: 1L metastatic PDAC
⏳ Outcomes:
• TFS 5.7 mo
• OS 11.2 mo (12.9 mo if meeting strict NAPOLI-3 criteria)
📊 DCR 57.5% | PR 37.5%
🧬 HRR-mutated pts: OS 16.2 mo vs 11.1 (p=0.043)
⚠️ KRAS G12D/V → worse OS
✅ Manageable grade 3–4 AEs (22.5%), lower than trials
💡 Takeaway: NALIRIFOX is effective, tolerable, & molecular subgroups matter. Routine HRR & KRAS profiling should guide therapy.

🚨 New in @ESMO_Open: Real-world analysis of NALIRIFOX in metastatic #PDAC confirms efficacy & tolerability!
https://t.co/ijdaqYz1bB
🎯 Median TFS 5.7m , OS 11.2m – consistent w/ NAPOLI 3
🧬 HRRm subgroup: OS 16.2 vs 11.1m
⚠️ KRAS G12D/V → worse OS (11.1m vs NR)
▪️ 22.5% G3–4 AEs
NALIRIFOX improves the curve, but for PDAC the summit is still far from reached ⛰️

ROME study @NatureMedicine randomized targeted therapy vs standard of care after molecular analysis (tissue/blood) in pretreated cancer patients across tumors. ⬆️ORR (17.5% vs 10%) & ⬆️PFS HR=0.66, but no OS (HR 0.92). OS feels important despite crossover ~45% . Congrats 🥳
1/3 Zenocutuzumab is @FDA approved for the treatment of NRG1-fusion-positive #cancers.
Knowing the frequency & distribution of NRG1-fusions is critical & this @Nature_NPJ#PrecisionOncology paper helps to get a better idea.

Was soll man sagen?
The results of SONIA are out on @Nature. 1L use of palbo did not improve PFS over 2L use, but led to more AEs & costs. An evolving treatment landscape challenges its interpretation— yet it shows that sequencing trials are possible. Congrats to the authors! https://t.co/XozXKro2sx
Out Now: The Breaking-ICE App!
https://t.co/8XQWIhdZ4g
ICE stands for Informative Censoring Exploration, and this app is designed to visually illustrate the impact of informative censoring on trial outcomes. Adjust parameters and watch as curves and statistical outputs change in real-time!
The aim? To demystify "informative censoring" and turn it from a dry statistical concept into a clear and intuitive idea, helping users understand why this phenomenon, when it occurs, can significantly bias trial results.
The App comes with a 4 minutes step-by-step video tutorial : https://t.co/ihd5oImaUQ. You can explore preloaded trials or perform your own sensitivity analyses by uploading your own data.
This project, funded by @hug_ge, and supported by the head of the Oncology Unit, @Alfdoc2, was inspired by the many works done with @VPrasadMDMPH and @AlysonHaslam within the @vkprasadlab ... stay tuned for the publication! Thanks 🙏
