Role of #neutrophil extracellular traps (#NETs) in #rheumatoid #arthritis (RA) pathogenesis
🔹RA neutrophils are prone to releasing NETs, where #citrullinated antigens (Cit-Ags), #carbamylated antigens (Car-Ags), and neutrophil #elastase (NE) are externalized.
🔹NET-cit- and Car-Ags are endocytosed by fibroblast-like #synoviocytes (FLS) and presented to antigen-specific T cells, leading to the generation of anti-citrullinated protein antibodies (#ACPAs) and anti-carbamylated protein (CarP) antibodies by B cells.
🔹Released NE cleaves peptidyl arginine deiminase (PAD)-2 from FLS. NE also degrades cartilage, and the resulting fragments are citrullinated by PAD-2 released from FLS.
🔹These antibodies can generate #immune complexes that, in turn, activate macrophages to release pro-inflammatory #cytokines.
🔹Carbamylated histones released within NETs trigger accelerated osteoclastogenesis after engaging TLR4. ACPAs, cytokines, rheumatoid factor and #immunocomplexes activate neutrophils to release NETs, creating a vicious cycle that can perpetuate #inflammation and joint damage.
