@AdrianoAguzzi Try reaching out to the Department of Biochemistry‘s technical workshop (@UZH). They have developed something similar for Rainin tips before. https://t.co/YsB8tZJF0d
@justindwalter Absolutely agree - cellular context step is crucial. Could be in later selection step or during screening. If one looks for an API. If binder needed for structural biology, needs and therefore strategies might of course differ.
@justindwalter I actually think it’s the other way around. It’s only now becoming technically feasible to do these campaigns with purified protein. There are currently lots of companies popping up around this particular approach in the GPCR space.
Today with @emblebi, we're launching the #AlphaFold Protein Structure Database, which offers the most complete and accurate picture of the human proteome, doubling humanity’s accumulated knowledge of high-accuracy human protein structures - for free: https://t.co/vtBGmTkKhy 1/
Nature article https://t.co/lHPFHHcpZD showing #GPCR isoform signalling bias, and a GPCRdb resource https://t.co/5FQMrhod0y. Another landmark study from @m_madan_babu group and great teamwork by Marti-Solano et al. Thanks to @Lundbeckfonden and @novonordiskfond
The key to obtaining the structure was boosting the expression levels of the receptor (which are almost non-existent for the wild-type) by #DirectedEvolution and further stabilizing the evolved receptor by protein engineering. (3/3)
Better late than never: We are happy to present the crystal structure of the human oxytocin receptor #GPCR in complex with the antagonist retosiban! Full story: https://t.co/1UbH4eRaRp (1/3)
Several cool findings: (i) we observe a cholesterol molecule bound to the receptor, which is known to be critical for OTR function and (ii) we located the elusive magnesium binding site, which is exclusive to the oxytocin/vasopressin receptor family! (2/3)
And… we're live! Our manuscript is out in @nature: 'Structural basis of the activation of a metabotropic GABA receptor'. A ‘movie’ of four GABA(B) heterodimer #cryoEM structures along its activation pathway. https://t.co/oEyFgNQwNQ #GPCR@SLAClab 1/n
New single-molecule FRET paper out! We study (on actual individual molecules) how the interaction between two disordered proteins is influenced by polymeric crowders (PEG). https://t.co/rymZwkIhTY (1/3)
Direct coupling to inward rectifying potassium channel Kir7.1 independent of G protein signaling & Ca2+ as a cofactor for both agonist/antagonist binding- great insights from MC4R crystal structure! Congratulations! #GPCR https://t.co/SasNEIuBxk
