I'm wildly proud to share our FIRST Weber Lab paper, out today in @Nature! I want to share a story about how this whirlwind of a project came together. A 🧵 on teamwork, open science, and how far we can go when we intentionally build a culture of trust in research. 1/
Memory genes are enriched in CAR T cells that persist and control tumors in patients. But which transcription factors drive beneficial memory programs?
We answer this in our lab’s first paper in @Nature led by @alexdoan96@kpmueller1@andy_yhchen. 1/

Our new, comprehensive review on CAR T cell persistence is officially published in @CellCellPress@trendscancer. Fantastic work from lead authors @kpmueller1@JGrenierMDPhD.

Our field has come a long way since the first CAR T success stories; still, reliable CAR T persistence remains elusive. Our goal is to highlight both lessons learned and open questions, which we hope will one day lead to consistently durable cures. Thanks for reading! /🧵
Durable CAR T persistence is among the great unsolved challenges within our field. But what do we actually mean when we say a CAR T cell is "persistent"? @JGrenierMDPhD, @EvanWeberPhD and I attempt to answer this question in our new review @trendscancer. 1/
Finally, we propose a unifying framework to define the concept of CAR T "persistence". We argue that the underlying mechanisms likely differ between active tumor killing vs. long-term immunosurveillance; we refer to this as "type I" and "type II" persistence, respectively. 5/
Congratulations to Dan Pham for her paper published today! Dan showed that autofluorescence lifetime imaging can monitor single cell metabolism to improve CAR T cell metabolic fitness. @sahakris@UWCarbone@Morgridge_Inst@UWMadEngr

There are days in life that shake you.
I’m shattered 💔 to share that I just found out that the US Government terminated my 2024 NIH Director’s Early Independence Award (~$2 million), threatening my long-promised assistant professor job at @Columbia & academic career... 1/🧵

Happy to report our work using #GBM organoids to predict #CART response in #glioblastoma@PennCancer
Excited to share our latest work in @NatureCancer to get a better understanding why relapsed T-ALL is so deadly and potential target strategies, led by Jason Xu and @changya1 with members of David Teachey, @junjyang, @CMullighan and many great colleagues. https://t.co/SaG7Q74FMS.
In this new Review, Caroline Diorio, David Teachey & Stephan Grupp discuss the progress made and remaining roadblocks with allogeneic CAR cell therapies for cancer: https://t.co/9CXeGnKa10
I'm honored to share that I have been awarded an Irvington Postdoctoral Fellowship from @CancerResearch! This funding will support my ongoing CAR T cell therapy research with @EvanWeberPhD at @ChildrensPhila. 1/
