Electron-light cavities called lacunae have long been observed inside sperm nuclei. Earlier studies suggested that they might contain proteasomes, but their molecular contents remained unresolved.
Kolata and colleagues specifically examined these cavities in cryo-FIB-milled human sperm by in situ cryo-electron tomography. The DNA-free lacunae were packed mostly with uncapped 20S proteasome cores; a small fraction carried PA200, and no nuclear 26S particles were detected. The clusters arise during the spermatid stage, as histones are replaced by protamines.
The advance is not that condensed sperm were thought incapable of protein degradation. It is the direct 3D identification of a long-observed structure as a proteasome-rich compartment. What these proteasomes degrade in mature sperm remains unknown.
