Temporal and spatial composition of the tumor microenvironment predicts response to immune checkpoint inhibition

Apart from the unique longitudinal on-treatment cohort of patients with #BreastCancer whose tumors are profiled by multiplexed imaging, this preprint features an open source algorithm - SpaceCat - for automated feature extraction from multiplexed images. Greater than 800 features are extracted from each MIBI image here correlating with patient / treatment outcomes.

Our study highlights the possible translation of RFA + ICI in pancreatic cancer treatment. RFA triggers DAMP release and innate immune response activation, reshaping the TME, sustaining T cell-mediated tumor killing, and driving tumor cells toward a more differentiated phenotype.

We found that:
1) RFA primes tumor-specific T cell responses that can be boosted by immunotherapy.
2) RFA + Immunotherapy reprograms tumor cells and sculpts TME towards an anti-tumor milieu.
3) HMGB1 and NLR increase after treatment associated with better prognosis in PC patients
