In this work, we find the PARP1-XRCC1/Lig3 axis, in collaboration with ATM, suppresses MMEJ. Inhibiting DDR components promotes genome-wide translocations while eliminating 53BP1 severely limits repair capacity of this alternative end joining pathway.
