SOX2, a key TF for transformation, is induced by TP53/RB1 inactivation, but downregulated by selinexor, thus hindering #lineageplasticity. Indeed, if we ectopically overexpress SOX2, we can rescue the transformation neuroendocrine phenotype.

Neuroendocrine transformation of adenocarcinomas occurs frequently upon treatment with targeted therapy, leading to poor prognosis. We have predictors of high risk of transformation (TP53/RB1 inactivation), but not therapies available to prevent this #lineageplasticity phenomenon
