Fantastic to see #mTurquoise2 expressed in monkey brains together with #mNeonGreen and #mRuby2

#mTurquoise2 has a 'relatively high QY' - the highest quantum yield among monomeric fluorescent proteins, to be precise.

A couple of words on #mTurquoise2 in a review of Quantum Biology: An Update and Perspective

There are a few 'blue' colonies that represent the original #mTurquoise2 template. We think that bright colonies reflect T203Y and dimmer colonies are T203H - sequencing will tell.....
Our first attempt at In Vivo Assembly (IVA) for site-directed mutagenesis: residue T203 was targeted in #mTurquoise2 and changed to Y or H - efficiency >90%

Huh? Same file openend in ImageJ (left) versus @FijiSc (right), showing #mTurquoise2 labeling of the endoplasmic reticulum

It turned out that tdTomato is a good FRET acceptor and we generated an EPAC sensor that uses #mTurquoise2 and tdTomato as a FRET pair. The Kd for cAMP is 4µM and a non-binding mutant (R279L) hardly responds

The original EPAC-based cAMP sensor uses a #mTurquoise2-Venus FRET pair. However, Venus is pH sensitive, which is problematic in yeast. Therefore, we were looking into acceptor with a low pKa

@mpbruchez @samuelconnell @TalleyJLambert @erin_rod_phd @MillerChemBio @NathanShaner @HenriquesLab The spectrally identical #mTurquoise2 has a slightly higher brightness in cells and a much better photostability.
The improved photostability is independently observed in other labs, but may depend on the power and illumination method.
Note the superior photostability of #mTurquoise2 relative to mCerulean3
This has been documented as well in our original study:

@timisstuck @NathanShaner If you want a tried and tested acid-resistant FP try mCherry, tagRFP(-T) or #mTurquoise2

@andrewng_synbio mEGFP would be my first choice: Truly monomeric and spectrally compatible with virtually any microscope.
Next to that I can recommend #mTurquoise2, mNeonGreen, mScarlet-I, mCherry (but check filters and excitation source first)
@wormsense @Vermeer_Lab @pedrosaurio #mTurquoise2, we've gotten pretty good results:
