Thrilled to share #PRIMARY2 Trial Results. This is the first randomised trial to show that adding [⁶⁸Ga]Ga-PSMA-11 PET-CT to MRI can safely halve the number of men needing a prostate biopsy.
Hot off the press in @TheLancetOncol 🎉
In 660 men across Australian sites with PI-RADS 2-3 but high clinical risk, PSMA-PET:
- Avoided biopsy in nearly half (49%)
- Was non-inferior for detecting clinically significant cancer (12% vs 16%)
- Halved the overdiagnosis of insignificant cancer (32% to 14%)
Fewer biopsies, less overdiagnosis, whilst not missing clinically significant prostate cancer: a real step forward for the diagnostic pathway.
Enormous congratulations to @ButeauJames@PeterMacCC , who led this as the centrepiece of his PhD and presented it as a plenary at #EAU26 in London. And to my brilliant co-lead @drlouiseemmett - this was a true partnership across our two centres.
Thank you to the UROLOGISTS @DrDanielMoon@declangmurphy (& many others), nuclear medicine physicians, technologists, radiopharmaceutical scientists across every site, and to @AnnetteVDHeyden and the @pros_tic who held it all together.
We are grateful to our funders: @PCF_Science, @nhmrc, St Vincent's Curran Foundation, Peter MacCallum Cancer Foundation, and @ANZUPtrials
Most of all, thank you to the 660 men who took part.

6/ In summary, HSD3B1 has 2 separate functions. First, it regulates cell-autonomous androgen biosynthesis that regulates prostate cancer outcomes and lethality. Second, it plays a tumor-extrinsic role in gut physiology and metabolism. Please read the paper! @PCF_Science@theNCI
1/ For 15 years we studied how slow vs fast forms of HSD3B1 work in prostate cancer, drive ADT resistance & lethality. Why is the fast #HSD3B1 so common? Does it do something other than cause lethal prostate cancer? YES! Here's the story @jclinicalinvest! https://t.co/MnQEjXA9A3
Excited to share results from the EQUIP study, just presented at @SNM_MI in Los Angeles. #SNMMI26
Last November, we installed the world's first @GEHealthCare Omni Total Body PET/CT - a 128cm long-axis field-of-view system with next-generation digital detectors. These are the first clinical results from anywhere in the world on this device.
The short version: better images, in a fraction of the time.
We enrolled 25 patients in a direct intra-individual comparison - same patient, same tracer injection, scanned first on a conventional PET/CT and then immediately on the Total Body system. Tracers included F-18 FDG, Ga-PSMA & DOTATATE, F-18 DCFPyL, and I-124.
Striking results:: 83% reduction in acquisition time (2.5 min vs 15 min) - with superior image quality, and significantly better sharpness and lesion conspicuity. Remarkably, in 68% of patients, reviewers identified small lesions on Total Body PET that would have been missed on conventional imaging.
Congratulations to Dr Alicia Corlett for an awesome oral presentation, Dr Cameron Pain, @pj_physics and all the co-investigators. Thanks to @Cancer_Research , @NIFAus, @unimelb and @PCF_Science for supporting this research. This was an investigator-initiated trial, funded by GE HealthCare. Hearty congrats to @neerajaiims on his concurrent #ASCO26 presentation and @NEJM paper on his TALAPRO-3 data on combination of Talazoparib+enzalutamide! @ASCO@PCF_Science https://t.co/DX3W4qFson
@declangmurphy discusses the PROTEUS trial #ASCO26 - no real concerns about ADT as a comparator in the control arm. Notably complete responses are really uncommon in both arms. @ASCO@PCF_Science
Mary-Ellen Taplin @DanaFarber delivers her PROTEUS presentation @ASCO#ASCO26 - with impressive effects on path CR and MFS! I’m looking forward to more forthcoming data as well! @PCF_Science
@ASCOPres Dr Eric Small delivers a powerful presidential talk — personal tribute to his partner Dr Amy Lin who passed this yr from a rare ovarian cancer. Oncology advancements needed — for every patient. Every cancer. Everywhere.

Eric Small’s @ASCOPres Address. Cancer is personal. It affects our families, friends, and neighbors. Translating science to clinics everywhere. #science@ucsfcc
@CaPsurvivorship@DanaFarber presents data on a crossover trial between enzalutamide and darolutamide for CNS adverse effects, showing clear differences between these drugs and based on chemistry that enables BBB penetration #ASCO26@ASCO@PCF_Science
JUST In: TALAPRO-3 published in @NEJM
Adding #talazoparib to enzalutamide/ADT
=>3-year rPFS: 77% vs 56% in HRR-deficient metastatic prostate cancer !
Looking forward to full presentation by @neerajaiims who keeps changing SOC, one trial at a time.

LBA1/Plenary #ASCO26: PROTEUS trial
Perioperative apalutamide + ADT vs. placebo + ADT with radical prostatectomy in HR localized or locally advanced PC
Our very own #DrMaryEllenTaplin from @DanaFarber_GU will take the stage at #ASCO26 and present phase 3 PROTEUS trial. This is a key multimodal treatment question and could shift the paradigm on how we think about systemic therapy in the perioperative setting. Dr Taplin has worked on countless Neoadjuvant Strategies in prostate cancer and it is rewarding to see her work materialize at the big stage !
