Meta-analyses with existing microglial datasets support the emerging microglial immunometabolism pathway as a therapeutic target in aging-related disorders. This study provides a human microglial transcriptome resource and reveals implications of age, sex and APOE genotype.
Integrated bulk and single cell analyses revealed known and novel microglial substates and showed enrichment of age-associated module genes in pro-inflammatory & DAM-like clusters. These harbored neurodegenerative disease risk genes including APOE & PLCG2.
We identified a novel microglial signature, characterized its expression in bulk tissue & single cell microglia transcriptomes. We uncovered co-expression modules associated with age, sex, & APOE4 enriched for lipid and carbohydrate metabolism genes in microglia.
