Let me explain.
I find that pictures speak a thousands words , so let me start off with this very nice picture on the web.
This picture showcases a lot of the biological paths but let’s focus on the left side. Several of these I have previously posted about, for example integrins, RTKs, GTP, RAS, K-RAS controlling PI3k pathway and MAPK pathway, etc. but look how all the “business” occurs at or near the cell membrane, where FLNA resides. From my posts we know that FLNA (and FLNA phosphorylation) interacts and signals a wide variety of known and well established moieties, from Integrins, to RTKs, RAS, GTP, Kinases, STAT3, FLNA-CT which translocates HIF into nucleus, calpains, etc…the list goes on and on. And we know that many cancer publications have spoken highly about the potential of FLNA as a therapeutic target, but those researchers have not found interventions to enable a cancer approach through FLNA.
IMHO Cassava has now found this very likely novel drug candidate for FLNA for cancer, it’s simufilam and other Cassava’s related drugs targeting FLNA fate, i.e. Cassava’s technology platform. I am convinced that simufilam targeting the FLNA phosphorylation will be extremely meaningful to cancer therapies in combination or as a mono therapy given the indirect activity against RAS (through potent FLNA phosphorylation reduction activity) and the feedback loop that I have postulated.
What’s even more exciting to me that unlike RAS targeted approaches that are prone to drug resistance through high rates of mutations of the RAS genes, FLNA target approach by-passes this drug resistance issues because of the criticality of FLNA signaling to the cell. So simufilam or other FLNA binders can be used potentially as a mono therapy or in combination with existing cancer drugs to increase their effectiveness or help these drugs overcome drug resistance as the University of Milan eloquently had shown with pituitary tumors using simufilam with octreotide.

The problem is that no pharma or other sponsor will do studies to “prove” efficacy given the economics and anti-competition of the industry, and this applies to other “anecdotal” cheap therapies, which there are a plethora of. if you look at the basic science expts and animal models with relevant dosing, and data collected, which is not cost onerous to do, one see‘s a vast amount of info that supports the mechanisms behind these interventions. The system establishing proof in healthcare is simply not pragmatic or feasible unfortunately.
Because it’s doing more than simply a modest reduction in BP. How do we know? it’s tied to the biology of how those reductions occurs, certainly worth risk given how widely it’s been used. Is it proven? of course not. But put pieces together and you get a compelling benefit/risk

Papers in the medical literature are being retracted at an alarming rate. The Arnold Foundation has funded and supported platforms like PubPeer, which have played a central role in driving those retractions.
An innocent person deserves the right to face their accuser directly. Instead, the response has been systematic censorship of anyone whose work threatens powerful financial interests—interests explicitly tied to figures like @BillAckman and his portfolio.

I see patterns that go far beyond the surface. The way certain attacks involving Epstein are being weaponized appears designed to reflect on an entire religion. Nothing here is coincidental. As human beings under God, we are called to seek the path of righteousness.
