An important limitation for serial killing is the low amount of perforin available and the lack of synergy between granzyme B and perforin. The γδ T cells that generated higher co-occurrence of granzyme B with perforin in the lytic vesicles correlated with longer killing series.
Interestingly, their lytic potential is higher on the 3rd/4th day rather than after a 14-day expansion which corresponds with the granzyme B expression peak.
