RootPath

RootPath is a pioneer in NextGen gene synthesis and SynBio solutions. With clonal, kb-long genes starting from $0.03/bp, we enable biology at scale.

@duribeb Yes, customer owns the IP of the sequences AND the data. We keep them confidential and wouldn't use them to do anything (like model training) other than serving your research :)

[RootPath @RootPathGx]

Today we are launching RootPath BinderVerse, a cloud lab for AI-designed binders.

Sequence in, binding data out in as few as 3 days, gene synthesis time included! Antibodies and Fab fragments made with CHO cells (TAT from 6 days) and CAR-Ts made with primary T cells (TAT from 10 days) can also be created from your binder sequences and tested.

Trustable data at lightning speed -- Learn more at https://t.co/kPOYlK8KkM

@RobertLKruse Thank you for the shout out! ❤️ Definitely let us know when you have a project on the horizon!

That that's our bread and butter. Actually we don't simply use pools straight up, since they carry too many synthesis errors. We use what we call 'Molecular Toothpick' to isolate error-free copy of oligo, hifi-amplify, then robustly assemble into longer constructs. Product: individually delivered, ensemble NGS QC-ed, complexity-tolerant gene fragments up to 3.5 kb starting from $0.02/bp (order thresholds apply), on par with some pooled products. Caveat: for these products we maximize cost-efficiency, not speed. See: https://t.co/BLytavBqVi

We can also express and purify target in house if they are not commercially available.

[RootPath @RootPathGx]

@devcayer Yep. We have a fast mode and high throughput mode. The latter uses pools and cuts the cost by about half, at the expense of longer turnaround time: 2.5 to 4 weeks depending on workflow, which is on par with industry standard. See screenshot:

@Gabogonzalez515 That’s a great question. We can use MS2 to do a quick test. Are you designing protein or RNA?

[RootPath @RootPathGx]

@vinnypeptide Yes you can absolutely ship the antigen or have us express and purify the antigen for you.

[RootPath @RootPathGx]

@SaligramaLab @iskander Thank you for the shout out! 😀

[RootPath @RootPathGx]

@brad_woolf Patient-derived T cell or target cells? If it’s the former it’s already very similar to what we offer. Our CAR-T workflow uses healthy donor-derived primary T cells. If it’s the latter it’s just the matter of establishing tumor-derived cell lines. Non trivial but doable.

[RootPath @RootPathGx]

@J33P4 We certainly can! Let’s talk.😀

[RootPath @RootPathGx]

@TensorTwerker Thanks for the comment! This is also a testament of how powerful AI-based protein design has become. When the hit rate was low, wet lab was definitely the bottleneck. Now that we can preload much of the work to GPUs, DNA synthesizers got a break :)

[RootPath @RootPathGx]

@brad_woolf Are you referring to the CAR-T service? If so, yes, any cell line that can be procured from ATCC can be used as target cell, optionally engineered with target transgenes. Let me know if I misunderstood the question.

@KevinKaichuang Just to throwing our hat in the ring 😉Happy to help!

We are excited to share another #MegabaseSpark submission, titled 'Bio-Based Petrochemicals' from @sean_wirt: "We plan to survey enzymes for activity on non-native substrates, enabling the construction of novel metabolic pathways for bio-based commodity chemicals traditionally derived from petroleum. This research supports the transition to a robust bioeconomy." Like and repost if you think this idea is interesting! #SynBio #SyntheticBiology #DNA
