In my several decades in science, few moments have been as seismic as when AlphaFold burst onto the scene (seeing the first structure of a K+ ion channel, from Rod MacKinnon, was up there too). We rushed to fold every protein we had been working on, that had no structure, and suddenly so many things made sense. AF, then AF2 and AF3, are daily workhorses in our business of protein engineering.
And I am beyond thrilled that my friend and collaborator David Baker has also received the prize. One of the projects I’m most proud of was a computational design collaboration with him to produce a resorufin fluorophore ligase, and it was miraculous when the crystal structure matched his computational design exactly (https://t.co/qYfPbonK9b). We use the tools David’s lab has produced, such as proteinMPNN and RFDiffusion, daily. As incredible as David’s contributions have been, I feel that his best work is ahead of him.
