Daraxonrasib shows a massive OS benefit in phase 3 RASolute 302.

👥 Study population:
Previously treated metastatic PDAC (RAS-mutant & WT)

⚔️ Trial arms:
🟣 Daraxonrasib 300 mg OD
🔵 Investigator’s choice chemotherapy

📊 Key results:
✔️ OS: 13.2 vs 6.7 months
✔️ HR 0.40 (p < 0.0001)
✔️ PFS also significantly improved

🛡️ Safety:
Well tolerated, no new safety signals

💡 Why this matters:
Post-progression options in PDAC have been weak.
A ~2x OS improvement is something we’ve almost never seen here.

⚠️ But:
Topline press release data only
Full breakdown (subgroups, ORR, QoL) awaited at ASCO

🔥 Takeaway:
If confirmed, RAS(ON) inhibition may redefine PDAC beyond KRAS G12C

Today’s KRAS win didn’t start today.
It took decades.

Daraxonrasib nearly doubled survival in pancreatic cancer. That’s extraordinary!

But lets zoom out for a sec:
▪️KRAS was called “undruggable.”
▫️NIH quietly poured ~$172M behind it.
▫️Funding grew 9x.
💡One insight (Shokat lab, 2013) changed everything.

This is how breakthroughs are supposed to work:
public money → basic insight
academic labs → first leap
industry → drug + survival curve

Today’s headline has MANY authors.

This ESTRO guideline outlines evidence-based recommendations for implementing IMRT and IGRT in #RectaclCancer radiotherapy☢️ . Supine positioning with a comfortably full bladder is preferred. Mandatory organs-at-risk include bladder, bowel cavity, and femoral heads. Auto-delineation and planning tools are supported with quality assurance. Accounting for setup uncertainties is essential. Daily volumetric image guidance is recommended to ensure accurate treatment delivery and minimize gastrointestinal and genitourinary toxicity. #RadOnc @ESTRO_RT
