So the contribution of deacetylation to whole cell acetyl-CoA metabolism appears limited 🥲Although the contribution may vary across subcellular compartments.
Variation in the extent of deacetylation between different histone lysine sites suggests selectivity in this response:
We didn't detect any release into extracellular acetate, so we checked 13C-labeling. We found that up to ~10% of the acetyl-CoA pool was derived from deacetylation, and this label was transmitted downstream into (iso)butyryl-CoA!
Our data are consistent with a model where acetate is preferentially mobilized to maintain other acetylation events (such as shown recently by Mendoza et al @berger_lab @GarciaLabMS)
